Single-stranded DNA-binding Proteins

چکیده
The emergence of DNA as the predominant hereditary material of life has necessitated the evolution of proteins whose sole function is the maintenance and care of these vital molecules. In a typical cell, there is a wide array of proteins with specialized functions in DNA metabolism that allow the cell to maintain and replicate its genome. In order to copy or make repairs to DNA, the double helix must be unwound to reveal the two complementary strands. The need to manipulate DNA in its singlestranded form has given rise to a specialized group of proteins. This article examines the family of proteinswhose primary role is to recognize and bind to single-stranded DNA(ssDNA).By comparing a variety of ssDNA-binding proteins from different species and different metabolic processes, we can discern their common characteristics, as well as the distinctions that make them essential to the cell.

ABSTRACT In eukaryote cells, DNA is complexed with a series of basic proteins making units of chromatin structure named nucleosomes. In addition, nonhistone proteins with different function are the components of chromatin. Among these proteins, a group with a low mobility on gel electrophoresis have been identified and named LMG. In this study a LMG protein with a molecular weigh of 160 ...
متن کاملTcaR–ssDNA complex crystal structure reveals new DNA binding mechanism of the MarR family proteins
The teicoplanin-associated locus regulator (TcaR) regulates gene expression of proteins on the intercellular adhesion (ica) locus involved in staphylococci poly-N-acetylglucosamine biosynthesis. The absence of TcaR increases poly-N-acetylglucosamine production and promotes biofilm formation. Until recently, the mechanism of multiple antibiotic resistance regulator family protein members, such a...
متن کاملE. coli SSB Activates N4 Virion RNA Polymerase Promoters by Stabilizing a DNA Hairpin Required for Promoter Recognition
Bacteriophage N4 virion RNA polymerase transcription of double-stranded promoter-containing DNAs requires supercoiled template and E. coli single-stranded DNA-binding protein (EcoSSB); other single-stranded DNA-binding proteins cannot substitute. The DNA determinants of virion RNA polymerase binding at the promoter comprise a small template-strand hairpin. The requirement for EcoSSB is surprisi...
متن کاملInteraction of bacteriophage T4 and T7 single-stranded DNA-binding proteins with DNA.
Bacteriophages T4 and T7 are well-studied model replication systems, which have allowed researchers to determine the roles of many proteins central to DNA replication, recombination and repair. Here we summarize and discuss the results from two recently developed single-molecule methods to determine the salt-dependent DNA-binding kinetics and thermodynamics of the single-stranded DNA (ssDNA)-bi...
متن کاملThe binding of in vitro synthesized adenovirus DNA binding protein to single-stranded DNA is stimulated by zinc ions.
We have synthesized wild type DNA binding protein (DBP) of adenovirus type 5 (Ad5) and several truncated forms of this protein by a combination of in vitro transcription and translation. The proteins obtained were tested for binding to a single-stranded DNA-cellulose column. It could be shown that for binding of in vitro synthesized wild type DBP, it is necessary to add zinc ions to the in vitr...
متن کاملIncrease of the DNA strand assimilation activity of recA protein by removal of the C terminus and structure-function studies of the resulting protein fragment.
A proteolytic fragment of recA protein, missing about 15% of the protein at the C terminus, was found to promote assimilation of homologous single-stranded DNA into duplex DNA more efficiently than intact recA protein. This difference was not found if Escherichia coli single-stranded DNA binding protein was present. The ATPase activity of both intact recA protein and the fragment was identical....
